I am traveling to Florida by car and wondering what time of day is best to travel in by car and with kids.

Dusk with kids, all by yourself would be noon.

If you can handle it, traveling by night is easiest as the children usually always end up falling asleep and then you can put in your own CD music and/or station and enjoy the quietness and your music group that you have been longing to listen to for a while now because the children are maybe constantly changing channels or interrupting the silence with arguments, etc.
It also seems to be harder to keep them occupied and busy without arguments along the way. A fun game that keeps everyone busy and not in tuned with their ipod, dvd movie, etc. which they can watch or use all of the time at home is the A-Z game. Look for the letter A for possibly the start of the word on signs, etc. Once that is mastered, then go to B and so on and so on until Z or you stop for a break.
You can do something similar with license plates, or my little eyes see something that is .... and have someone guess what it is. Once the person announces the correct answer, then it is their turn. This way, they can see the scenery of the area along the way and what is beyond the city in which they live.

That is a very personal choice. I have done my share of driving by myself. I would rather leave early early in the morning to get most of the driving done in the daylight. With kids is another game. Restroom stops I guess increases due to smaller bladders. To your trip add up like 10 min to 15 min with each stop every 2 hrs. Day time is always safer.
